miRNA and Gene Expression

The Genomics Shared Resource provides miRNA and gene expression analysis on several different platforms depending on the organism, starting material and size of your project. The GSR offers genome wide expression analysis with small RNA Seq and microRNA Seq and targeted analysis using NanoString and Taqman.

NGS SmallRNA Sequencing : TruSeq Small RNA Library Preparation kits generate small RNA libraries directly from total RNAs.  MircoRNAs generated by Dicer processing are efficiently targeted by the included modified adapters.  With small RNA-Seq you can discover novel miRNAs and other small noncoding RNAs and examine differential expression of all small RNAs in any sample.

NanoString nCounter Gene Expression

  • Directly assay serum/FFPE
  • Human Cancer Reference Panel- Gene Expression CodeSet profiling 230 cancer-related human genes and 6 internal reference genes.

TaqMan assays

  • Gene expression: > 1M TaqMan gene expression assays available for RT-PCR using the QuantStudio 6 Flex Real-Time PCR System.
  • miRNA expression: TaqMan miRNA assays available for quantitative RT-PCR using the QuantStudio 6 Flex Real-Time PCR System.

RNA- Generally, 1 µg total RNA is required per Agilent, Exiqon or Illumina gene expression or miRNA array. Note: It is recommended to treat RNA samples with DNase I. All RNA samples must have a A260/280 between 1.8 - 2.1, A260/230 > 1.6 and a Bioanalyzer RIN > 7.0. It is required that the user provide the concentration, volume, buffer and preparation method of the sample. Degraded RNA samples can be analyzed (i.e. paraffin, formalin fixed tissues) with Exiqon RT-PCR miRNA assays.
