Microarray and Genomics Facility
Description
The Microarray and Genomics Facility will afford RPCI investigators a full spectrum of services for parallel analyses of genomes and their expression. Applications of this technology include large scale SNP genotyping and methylation (Sequenom MassARRAY and Illumina GoldenGate and Infinium), BAC and oligo array based Comparative Genomic Hybridization (aCGH), and global gene expression profiling (Agilent and Illumina arrays). Variations of these applications can be utilized to: detect and map chromosomal aberrations, loss of heterozygosity and SNPs, profile gene expression patterns, identify differences between two populations of DNA or RNA (ie. tumor vs normal, treated vs untreated cell populations), determine the effect of therapeutic agents on the genome and their effect on gene expression, and determine the effect of genetic alterations on gene expression patterns in transgenic and knockout mice. The facility also maintains and provides clone distribution services for fifteen large insert genomic BAC/PAC libraries for human, mouse, rat, and S. Pombe as a resource for RPCI investigators. In addition, investigators with high throughput projects are supported by the robotics infrastructure within the facility.

Using the Facility
Investigators should view the instructions and policies described. Contact the facility (Jeff Conroy 716-845-3361) for additional information regarding use of the services and requisition forms. The array facility is housed in a clean room environment in order to produce high quality arrays. Personnel access is minimized to reduce dust and traffic around the arrayer. Unless scheduled for training or approved for access, non-microarray personnel are not permitted in the array facility. Please call or email with any questions related to this request.

Services
Investigators can request genotyping and microarray services including BAC array CGH, large scale SNP detection, SNP analysis (Illumina and Sequenom), copy number determination, quantitative gene expression, methylation studies, RNA/DNA isolation, Cy-3 & Cy-5 probe generation, hybridizations, custom-made cDNA, oligo and genomic BAC arrays, Agilent and Illumina arrays, scanning of microarrayed slides, image analysis, normalization of data and web-based interface with Oracle relational database. In addition to the cDNA libraries, fifteen BAC/PAC genomic libraries are available for clone selection, characterization, mapping, and distribution.
Scheduling
Investigators should contact Jeff Conroy (716-845-3361) for details. Priority is given to NCI-funded investigators with RPCI affiliation, and as determined by the RPCI Microarray Steering Committee.
User Fees
There is a chargeback system for users of the RPCI DNA Microarray and Genomics Facility. The fees vary depending on the affiliation of the investigator, cost of materials and the extent of services requested from the user (fee schedule).
Billing Procedure
Requisition forms must be completed, including purchase order/account number and signature, and submitted along with the sample(s) and signed MTA (if required). Billing is done at the completion of the service.
